- Zanidatamab, a HER2-targeted bispecific antibody, was evaluated with and without tislelizumab (anti-PD-1) plus chemotherapy vs. trastuzumab plus chemotherapy in HER2-positive gastroesophageal adenocarcinoma.
- In a phase 3 trial, zanidatamab-tislelizumab-chemotherapy and zanidatamab-chemotherapy both showed improved progression-free survival (median 12.4 months vs. 8.1 months with trastuzumab-chemotherapy).
- Zanidatamab-tislelizumab-chemotherapy demonstrated longer overall survival (median 26.4 vs. 19.2 months) compared to trastuzumab-chemotherapy, while zanidatamab-chemotherapy showed a non-significant improvement in overall survival at interim analysis.
- Grade 3 or higher adverse events occurred in 83.3% (zanidatamab-tislelizumab-chemotherapy), 73.8% (zanidatamab-chemotherapy), and 74.5% (trastuzumab-chemotherapy) of patients, with diarrhea being the most common severe adverse event.
