  • Adamtsl3, a schizophrenia-associated glycoprotein, is a cell-autonomous regulator of perineuronal net (PNN) integrity in parvalbumin-positive (PV+) interneurons.
  • Deletion of Adamtsl3, either postnatally or in adulthood, leads to PNN deficits by increasing MMP9 activity, reducing PNNs, decreasing Otx2 uptake, and raising oxidative stress in PV+ cells, which can be reversed with MMP9 inhibition.
  • Conditional Adamtsl3 deletion in adult PV+ interneurons reactivated juvenile-like ocular dominance plasticity, highlighting its role in adult cortical plasticity and schizophrenia pathophysiology.
