- ABBV-706 is a SEZ6-targeting antibody-drug conjugate with a topoisomerase-1 inhibitor, studied in a phase 1 trial for advanced solid tumors, including relapsed/refractory small cell lung cancer.
- The trial included 288 patients, with 240 receiving monotherapy; the most common treatment-related adverse events were anemia (61%) and fatigue (38%), with grade 3 or higher events occurring in 61% of patients.
- In the relapsed/refractory SCLC monotherapy cohort of 124 patients, the objective response rate was 52%, showing promising efficacy, with similar response rates at doses of 1.8 mg/kg and 2.5 mg/kg.
- Based on safety, efficacy, and pharmacokinetic data, the recommended phase 2 dose was determined to be 1.8 mg/kg every 3 weeks, due to a better safety profile and highest overall survival of 12.4 months in SCLC patients.
- The study highlights ABBV-706 as a potential treatment option for advanced SCLC, warranting further investigation in phase 2 trials.
