Safety and efficacy of concurrent radiation therapy with amivantamab in patients with advanced EGFR- or MET-altered non-small cell lung cancer - PubMed

  • Amivantamab concurrent with radiation therapy (RT) was studied in advanced EGFR- or MET-altered NSCLC patients.
  • The retrospective analysis included 14 patients receiving 21 RT courses, mostly for palliation (71%) or oligoprogression (29%).
  • Common mutations were EGFR ex19del (64%), L858R (14%), ex20ins (14%), and MET exon 14 skipping (7%).
  • RT-related adverse events were mild: 13 grade 1 acute, 6 grade 2 acute, and 3 grade 1 late events; no grade ≥3 AEs occurred.
  • Symptom relief was achieved in 87% of palliative irradiations, and local control was maintained in 80% of cases.
  • Median follow-up was 8.0 months for AEs and 9.5 months for oncologic outcomes.
  • The 6- and 12-month overall survival rates were 77% and 50%, respectively.
  • For patients continuing amivantamab post-RT, median PFS was 4.5 months, TTD 4.8 months, and TTNT 5.5 months.
  • Concurrent palliative RT with amivantamab appears feasible and well-tolerated, warranting further validation.
