- Elecoglipron (AZD5004) is an oral, once-daily small molecule GLP-1 receptor agonist without food restrictions, being developed for obesity and overweight.
- A phase 2, double-blind, randomized, placebo-controlled trial (VISTA) in adults with obesity/overweight without type 2 diabetes, from multiple countries.
- 310 participants were randomized to elecoglipron doses (5, 15, 50, 75 mg with different titration schedules) or placebo over 36 weeks.
- At 26 weeks, weight change ranged from -2.6% (5 mg) to -10.5% (75 mg weekly titration) vs -0.6% placebo; ≥5% weight loss: 40.4–88.8% vs 15.6% placebo.
- Adverse events (mostly nausea, constipation, diarrhea, headache, vomiting) occurred in 84–98% of elecoglipron groups vs 84% placebo, with tolerability consistent with GLP-1 agonists.
- The study supports phase 3 investigation for weight management, funded by AstraZeneca.
