- Co-cultured human brain microvascular endothelial cells and pericytes with hBOs under dynamic flow, which enhanced BBB function (increased TEER, reduced permeability).
- Induced parenchymal pathology with Aβ42 oligomers in hBOs, leading to neurite loss, impaired proliferation, apoptosis, and cytokine upregulation.
- Pathological hBOs disrupted BBB integrity, decreasing tight junction protein expression and increasing permeability.
- The model shows physiological hBOs strengthen BBB, while pathological ones impair it, useful for studying neurological disorders and drug discovery.
