- The HER2×CD2 bispecific provides tumor-dependent costimulation, enhancing TCE efficacy and reducing toxicity.
- It restores anti-tumor activity at low TCE doses, preventing TCE-driven toxicity while maintaining effectiveness.
- In a xenograft model, co-treatment with HER2×CD2 led to complete tumor remission in 8 of 9 mice, compared to 1 of 9 with TCE alone.
- HER2×CD2 compensates for loss of CD58 on tumor cells, a common immune escape mechanism.
- Unlike CD28-based costimulation, it activates CD28-negative CD8 T cells, important in elderly patients and solid tumors.
