- Sanggenol L (SL) exhibits potent inhibitory effects on glioblastoma with minimal toxicity to normal cells.
- SL induces cytotoxic endoplasmic reticulum (ER) stress, suppresses cell viability, and triggers apoptosis in glioblastoma cells.
- Mechanistically, SL binds to BiP, blocking its interaction with IRE1α, leading to IRE1α dimerization and activation of the IRE1/CHOP/XBP1s/Bcl-2 pathway.
- SL also promotes ubiquitination and degradation of MGMT, enhancing glioblastoma chemosensitivity to temozolomide.
