- Lenvatinib combined with pembrolizumab did not improve overall survival compared to standard of care in patients with recurrent/metastatic head and neck squamous cell carcinoma in the phase 2 LEAP-009 trial.
- Median overall survival was 8.4 months for lenvatinib-pembrolizumab versus 11.3 months for standard of care, with a hazard ratio of 1.48 favoring standard of care.
- Treatment-related adverse events led to higher discontinuation rates with lenvatinib-pembrolizumab (16.8%) and lenvatinib monotherapy (15.9%) compared to standard of care (8.6%), and there were more treatment-related deaths in the lenvatinib-containing arms.
- The study highlights the need for further research to identify effective second-line treatments for this patient population after progression on first-line platinum-based therapy and PD-(L)1 inhibitors.
