  • Neutrophil extracellular traps (NETs) worsen lung injury by inducing pyroptosis in alveolar macrophages (AMs).
  • Gasdermin D (GSDMD)-mediated pyroptosis is a key factor in chronic obstructive pulmonary disease (COPD) progression.
  • Mice exposed to cigarette smoke (CS) for 16 weeks developed COPD-like symptoms, with NETs increasing inflammation and lung damage.
  • NETs also increased reactive oxygen species (ROS), promoting GSDMD deubiquitination and pyroptosis in AMs.
  • Treatment with DNase1 (to degrade NETs) or GSDMD knockdown reduced pyroptosis, inflammation, and improved lung function.
  • Targeting GSDMD or NETs could be a new therapeutic strategy for COPD.
