- The randomized phase 2 trial (HCHTOG1906) compared sequential versus concurrent neoadjuvant immunochemotherapy in locally advanced esophageal squamous cell carcinoma.
- Patients received either sequential administration (chemotherapy on day 1, toripalimab on day 3) or concurrent administration (all drugs on day 1).
- No significant difference in pathological complete response (pCR) rates was found between sequential (17.8%) and concurrent (26.9%) groups.
- Concurrent treatment was associated with higher incidences of nausea and diarrhea, and more treatment-related deaths (5 vs. 1).
- Overall survival and disease-free survival did not differ significantly between the two groups.
