- The Brake-Drive Osteo System is a biomaterial designed to sequentially address chronic inflammation and impaired osteogenesis in osteoporotic bone defects.
- It consists of a calcium phosphate scaffold (BCP-T/N@Q/V) integrating quercetin-loaded nanovesicles to first reduce inflammation and teriparatide-loaded nucleic acid frameworks to then promote bone regeneration.
- The system reprograms macrophages to a pro-regenerative phenotype, mitigates inflammatory stress, and enhances osteogenic maturation of osteoporotic bone marrow mesenchymal stem cells.
- In a rat osteoporotic femoral condyle defect model, it achieved accelerated and structurally integrated bone regeneration, demonstrating translational potential.
