- The EMPHASIS trial evaluated the efficacy and safety of minocycline in patients with acute ischaemic stroke.
- A multicentre, double-blind, randomised, placebo-controlled trial was conducted across 58 hospitals in China.
- Patients received minocycline or placebo within 72 hours of stroke onset, with NIHSS scores between 4 and 25.
- Primary outcome was excellent functional outcome (mRS score 0-1) at 90 days, favoring minocycline (52.6% vs. 47.4%).
- Minocycline showed a significant benefit in functional outcomes without increased safety concerns.
- No significant differences in symptomatic intracranial haemorrhage or serious adverse events were observed between groups.
- Future studies are needed to confirm benefits in patients with more severe or minor strokes.
