- Four rare cases of early-onset tau-independent β-amyloidosis were found among 397 banked brains from donors with hematological malignancies or cardiovascular diseases.
- In the study, 17 brains from blood cancer patients and 3 from cardiovascular disease patients were examined, revealing Aβ plaques in adult brains without tau pathology.
- Vascular injuries were present in all 17 blood cancer brains, with iron leakage in 13/17 and axonal pathology in 13/17.
- Malignant cell infiltration occurred in 5/14 blood cancer brains, and light chain infiltration was found in all 3 multiple myeloma cases.
- In cardiovascular disease brains, Aβ deposition as diffuse plaques was seen in the cerebral cortex, with vascular and axonal pathologies in white matter regions.
- Multi-labeling showed injury-induced axonal pathology was linked to β-amyloid processor protein and β-secretase 1 upregulation, but not intra-axonal amyloid tracer staining.
