  • Parathyroid hormone-related protein (PTHrP) is identified as a therapeutic target in idiopathic pulmonary fibrosis (IPF).
  • PTHLH, the gene encoding PTHrP, is upregulated in IPF lung tissue, especially in bronchial epithelial cells.
  • PTHrP1-34, a processed form of PTHrP, is elevated in fibrotic lungs and activates fibroblasts, leading to extracellular matrix production and fibrosis progression.
  • Neutralizing antibodies, peptides, and gene silencing targeting the PTHrP1-34/parathyroid hormone 1 receptor axis showed attenuating effects in a bleomycin-induced pulmonary fibrosis mouse model.
  • The study suggests that PTHrP1-34 from bronchial epithelial cells plays a key role in IPF pathogenesis and is a promising therapeutic target.
