- Developed a novel method for expanding murine regulatory T-cells (Tregs) from wild-type mice without genetic modifications, ensuring high Foxp3 purity (>99%).
- Achieved robust Treg expansion (over 30-fold) while maintaining Foxp3 expression and high levels of CD25 and CD62L, using anti-CD3/CD28 bead activation and human IL-2 over 7 days.
- Enabled efficient retroviral transduction (over 90% efficiency) on day 3 without compromising expansion, producing CAR Tregs that effectively target CD19+ cells and retain suppressive activity.
- Demonstrated in vitro and in vivo functionality, including protection against acute graft-versus-host disease, supporting use for preclinical CAR Treg development.
