  • Hydrogen gas (H2) pretreatment via intraperitoneal injection improves acute liver injury induced by lipopolysaccharide (LPS) in mice.
  • H2 reduces inflammatory cell infiltration, lowers serum ALT and AST levels, and modulates oxidative stress markers (decreases 3-nitrotyrosine, MDA, MPO; increases GSH).
  • Mechanistically, H2 inhibits TLR4-mediated signaling pathways (IKK-NF-κB and MAPK), leading to reduced pro-inflammatory cytokines (TNF-α, IL-1β, IL-18).
  • H2 suppresses NLRP3 inflammasome activation (NLRP3, ASC, Caspase-1) and pyroptosis signaling involving GSDMD, Caspase-8/11, Caspase-3, and GSDME, as shown by reduced TUNEL staining.
