  • Liver-specific STEAP4 knockout (L-STEAP4 KO) mice showed delayed liver recovery at 48 hours post-APAP, despite similar initial injury levels.
  • Loss of STEAP4 led to endolysosomal iron buildup, lysosomal membrane damage, and impaired mitophagy, reducing hepatocyte proliferation.
  • Deferiprone (DFP) restored mitochondrial and lysosomal functions, protecting against AILI.
  • Overexpressing STEAP4 or TFEB, or posttreatment with DFP, repaired lysosomal membranes and mitigated AILI.
  • Targeting STEAP4-mediated endolysosomal iron overload could offer new therapeutic strategies for AILI.
