- Diabetes mellitus (DM) is hypothesized to increase the risk of Alzheimer's disease (AD), but existing studies have shown conflicting results.
- This meta-analysis systematically evaluated the association between DM and AD risk, including 11 studies with 3,393,545 participants.
- Results showed DM significantly increases AD risk (HR = 1.36, 95% CI: 1.19-1.55, P < 0.00001), consistent across different sample sizes and populations (Asian and non-Asian).
- Adjusting for APOE ε4 mutations removed the statistical significance of the association (HR = 1.07, 95% CI: 0.97-1.19, P = 0.177).
- The study concludes DM is an independent risk factor for AD, but methodological limitations suggest the need for further high-quality prospective cohort studies.
