Ameloblastoma (AB) is a benign but locally aggressive odontogenic tumor with high recurrence and potential for metastasis.
Primary cilia (PC), usually reduced in malignancies, were consistently found in all AB cases but absent in oral squamous cell carcinoma (OSCC).
PC density varied among AB histological subtypes, with polarized orientation in follicular subtype peripheral cells.
PC were present in recurrent and metastatic AB lesions, though their role in progression is unclear.
Activation of the Hedgehog (Hh) signaling pathway in ABs was indicated by elevated GLI1 and SHH expression.
CEP164, a gene related to early ciliogenesis, was selectively upregulated in ABs and promoted PC formation.
CEP164 overexpression was localized near γ-tubulin, aligning with its role in basal body distal appendages.
CEP164 knockdown suppressed ciliogenesis, suggesting its key role in maintaining PC in ABs.
The study highlights CEP164 as a potential diagnostic and therapeutic target in AB.
