- Breast cancer (BC) patients have a higher prevalence of atrial fibrillation (AF) even before treatment compared to healthy controls.
- BC patients and mice models showed increased atrial remodeling and AF susceptibility, linked to elevated levels of soluble ADAM10 (sADAM10).
- sADAM10 promotes cleavage of Ephrin B2 into soluble Ephrin B2 (sEphrin B2), activating atrial fibrosis pathways.
- Inhibition of sADAM10 reduced atrial fibrosis and AF susceptibility in BC mice, suggesting a potential therapeutic target.
- The study highlights a direct role of BC in AF pathogenesis, independent of cancer treatment-related cardiotoxicity.
