  • MyD88 inhibitory Peptide-2 (MIP2) improves neurological outcomes and reduces neuroinflammation following intracerebral hemorrhage (ICH)-mediated secondary brain injury.
  • In a collagenase-induced ICH model, MIP2 treatment at days 0, 1, and 3 post-ICH improved neurobehavioral function, reduced hematoma volume, and enhanced blood-brain barrier integrity by upregulating tight junction proteins and lowering MMP9 expression.
  • MIP2 reduced neuroinflammation by suppressing glial cell activation, neutrophil infiltration, and inflammatory mediators such as HO-1, MyD88, NF-κB, TLR4, and IL-1β.
  • The peptide increased expression of anti-oxidative stress markers (GPX4, AIFM2, and Nrf2), likely through modulation of the MyD88-mediated signaling pathway.
  • MIP2 provides neuroprotection by preserving BBB integrity, suppressing apoptosis, and attenuating neuroinflammation while enhancing antioxidative defense after ICH.
