- Engineered natural killer (eNK) cells derived from human-induced pluripotent stem cells (iPSCs) and armed with five genes (CCL19, CCR2B, CD16, IL-15, NKG2D-DAP10) to enhance homing, persistence, and cytotoxicity.
- In vitro, eNK cells demonstrated significant long-lasting cytotoxicity and antibody-dependent cell-mediated cytotoxicity (ADCC) against human lung cancer cell lines.
- In orthotopic and subcutaneous cell line-derived xenograft models, eNK achieved near-complete tumor regression.
- In patient-derived xenograft models, eNK monotherapy modestly inhibited tumor growth (28% reduction), but combining with cetuximab enhanced efficacy (53% inhibition) via ADCC.
- eNK cells are positioned as a novel 'off-the-shelf' allogeneic therapy with potential for treating refractory solid tumors like lung cancer.
