- Bystander memory CD8+ T cells (TBYS) in tumors recognize pathogen antigens, are abundant and polyfunctional, unlike scarce exhausted tumor-specific CD8+ T cells.
- TBYS cells are heterogeneous, comprising TCM, TEM, and TRM subsets with distinct chromatin and transcriptional profiles, following a TCM→TEM→TRM differentiation trajectory during tumor progression.
- TRM cells show superior tissue retention and dominate the TBYS pool, and their differentiation is instructed by TGF-β from regulatory CD4+ T cells, which promotes accumulation by suppressing KLF2.
