- Narcolepsy type 1 (NT1) is hypothesized to be an autoimmune disorder linked to the loss of hypocretin (Hcrt; orexin) in the hypothalamus.
- Postmortem studies of NT1 brains revealed an 11-fold increase in CD4+ T-cells specifically in the Hcrt region, with no significant rise in CD8+ T-cells.
- The increase in CD4+ T-cells was localized to the Hcrt region, with no changes observed in other hypothalamic or extra-hypothalamic areas.
- CD4+ T-cells in the Hcrt region expressed tissue-resident memory markers CD49a and CXCR6, supporting the autoimmune hypothesis of NT1.
