- Immune cell profiling identified expanded stem cell-like memory T cells (TSCM) in anti-GAD65-associated neurological syndromes (AINS).
- Single-cell RNA and immune repertoire sequencing revealed higher frequencies of TSCM in peripheral blood mononuclear cells (PBMCs) and cerebrospinal fluid (CSF) of anti-GAD65 AINS patients.
- Clonally expanded CD4+ TSCM in CSF exhibited increased proinflammatory gene expression.
- Histological analysis confirmed the presence of intraparenchymal CD8+ TSCM and rare CD4+ TSCM in brain tissue of anti-GAD65 AINS patients.
- 25% of CSF B cell receptors (BCRs) were GAD65-reactive, showing increased somatic hypermutations compared to non-GAD65-reactive BCRs.
