  • Fibroblasts play a critical role in autoimmune diseases like rheumatoid arthritis (RA) by producing inflammatory mediators in response to TNFα.
  • Peptidylarginine deiminases (PADs) convert peptidylarginine to peptidylcitrulline, a process linked to RA and interstitial lung disease (ILD).
  • PAD2 transcript levels increase with age in lung fibroblasts (LFs) but not in fibroblasts from other organs.
  • PAD2 deficiency reduces expression of Podoplanin and TNFRSF1b in fibroblasts across multiple organs.
  • PAD2 deficiency attenuates TNFα-induced expression of LIF-dependent and LIF-independent genes, including IL-6, in LFs.
  • PAD2 regulates TNFα-induced gene expression in fibroblasts in an organ-specific manner, suggesting a novel pathogenic role in RA.
  • The findings provide a plausible explanation for the lung-synovium connection observed in RA.
