- CXCR4 mRNA overexpression is linked to poor survival in primary colorectal cancer but indicates better survival in metastatic tumors and predicts improved response to immune checkpoint inhibitors (ICB).
- Analysis of 15,026 CRC samples showed higher CXCR4 expression in metastatic sites than primary tumors and in non-liver vs. liver metastases, correlating with high TMB, dMMR, and PD-L1 positivity.
- High CXCR4 expression was associated with increased infiltration of B cells and M1/M2 macrophages in the tumor microenvironment, suggesting a role in immune modulation and ICB selection.
