  • Shared genetic architecture between sleep traits (sleep apnoea, insomnia, daytime sleepiness, snoring) and cardiometabolic diseases (CMDs) was investigated.
  • Significant genetic correlations were found in 25 out of 28 pairs of sleep traits and CMDs.
  • 754 independent SNPs (691 unique) and 102 colocalized loci (85 unique) were identified, with 47 SNPs (44 unique) validated.
  • Notably, rs429358 (APOE) showed pleiotropic effects across sleep apnoea, insomnia, and type 2 diabetes (T2D).
  • 48 annotated genes were validated, with shared genes enriched in mortality and growth-related phenotypes.
  • Pathway analysis highlighted Cushing syndrome, hormone secretion, and cGMP-PKG, Ras, and calcium signalling pathways.
  • Genetically predicted T2D increased risk of sleep apnoea, sleepiness, and snoring, while sleep apnoea was positively associated with heart failure and T2D.
  • The study suggests shared genetic determinants may explain epidemiologic associations and identifies potential treatment targets.
