Antiviral activities of multiple antivirals against highly pathogenic avian influenza A H5N1 in vitro and in mice - PubMed

  • The study evaluated antiviral effects of baloxavir acid (BXA), oseltamivir carboxylate (OSC), EIDD-1931 (NHC), and ribavirin (RBV) against five H5N1 strains in vitro, with BXA showing the lowest EC50.
  • Combination treatments (BXA/NHC and BXA/OSC) demonstrated more potent inhibitory effects than monotherapies in vitro.
  • In BALB/c mice infected with bovine H5N1 virus, 7-day treatments with BXM (baloxavir marboxil) alone or in combination with MNP (molnupiravir) or OSP (oseltamivir phosphate) increased survival rates to 50% and better inhibited viral replication compared to 5-day treatments.
  • Mice treated with combination therapies (BXM/MNP or BXM/OSP) had lower viral yields in lungs than those treated with BXM alone, and most mice died from severe neurological symptoms.
  • The findings suggest extending 7-day combination antiviral treatment should be considered for human H5N1 infection management.
