- Lung adenocarcinoma (LUAD)-derived leptomeningeal metastases (LM) are a major subtype of LM cases.
- Single-cell RNA sequencing of CSF cells from LUAD-LM patients revealed an immunosuppressive state with pro-tumoral macrophages and proliferating CSF circulating tumor cells (CSF-CTC).
- CSF-CTC enhances immunosuppression via the NECTIN2_TIGIT axis with CD8+T/NK cells and the CD47_SIRPA axis with antigen-presenting cells.
- Trophoblast cell surface antigen 2 (TROP2) levels in CSF are significantly higher in LUAD-LM patients compared to normal controls and LUAD patients without LM.
