  • Metoclopramide is commonly used for nausea and vomiting in pregnancy (NVP) and hyperemesis gravidarum.
  • The study investigates if metoclopramide-induced depressive symptoms are more frequent in pregnant women compared to non-pregnant women.
  • A disproportionality analysis was conducted using the Netherlands' national spontaneous ADR reporting system.
  • Reports showed depressive symptoms were more common in pregnant women (21.1%) than non-pregnant women (3.0%) (ROR 8.6; 95% CI 3.2-23.2).
  • Depressive symptoms were also more frequent with metoclopramide compared to other anti-emetics (7.3%) (ROR 3.4; 95% CI 1.2-9.3).
  • 33.3% of pregnancy-related cases reported suicidality tendencies alongside depressive symptoms.
  • Depressive symptoms were often accompanied by anxiety, panic attacks, agitation, and restlessness.
  • The study suggests pregnancy-related factors and metoclopramide's pharmacological mechanism may contribute to increased depressive symptoms.
  • Awareness of this ADR is crucial for early recognition and intervention to improve maternal safety.
