  • Plant retrograde transport is mediated by a novel class of spherical vesicles (30-50 nm) derived from multivesicular bodies (MVBs).

  • Arabidopsis SNX1 forms shorter membrane tubules than mammalian SNX1, and SNX1-SNX2 heterodimers can generate spherical vesicles similar to those observed in vivo.
  • Knockdown of SNX1 or SNX2 leads to vacuolar mislocalization and increased degradation of GFP-VSR2, impairing retrograde transport.
  • Defects in SNX1 and VPS29 inhibit the formation of these spherical vesicles, causing embryonic lethality before the globular stage in Arabidopsis.
