Shear stress activates Piezo1 signaling, which induces CD99L2 expression in primitive erythrocytes during a specific developmental window.
CD99L2 is essential for erythrocytes to detach from the endothelium and enter the bloodstream, as its loss leads to erythrocyte retention, impaired maturation, and hemolytic anemia.
Mechanistically, CD99L2 binds and anchors β-catenin at the membrane, preventing its nuclear translocation and the subsequent activation of Rap1 signaling and adhesion molecule expression.
This pathway, conserved in zebrafish and mouse models, couples hemodynamic forces to erythrocyte adhesion control, facilitating the initiation of blood circulation.
