Macrophage-rich niches regulate T cell dynamics at the liver invasive margin during gallbladder cancer progression - PubMed

  • Liver invasion is a common event in gallbladder cancer (GBC) progression.
  • Single-cell and spatial transcriptomics revealed CXCL9+ macrophage-rich immune cell niches at the tumor-liver invasive margin.
  • Two subclasses of CXCL9+ niches were identified: CXCL9+TRAC+ (CT) and CXCL9+C1QB+ (CC).
  • CD8+ T cells were recruited by CXCL9+ macrophages via CXCL9-CXCR3 interaction in the CT niche near the liver.
  • The CC niche, near the tumor core, induced CD8+ T cell exhaustion via LGALS4 expression.
  • High CXCL9 and low LGALS4 levels in the liver invasion margin correlated with better prognosis and anti-PD-1 immunotherapy response in GBC patients.
