- Early detection of urothelial carcinoma (UCa) is challenging due to limitations of current diagnostic methods.
- A novel urine-based H4C6/TWIST1 methylation assay was evaluated for diagnostic accuracy in UCa.
- The study analyzed 743 urine samples (387 UCa patients and 356 controls) from four hospitals.
- DNA was extracted from 3.5 mL of morning urine, and methylation status was assessed via quantitative PCR.
- The assay showed high diagnostic accuracy: 93.02% sensitivity and 92.13% specificity.
- Performance was strong for high-grade (97.64%) and muscle-invasive tumors (97.50%).
- Diagnostic efficacy was consistent across age, sex, and tumor location (lower and upper urinary tract).
