Diagnostic performance analysis of two-gene methylation in urothelial carcinoma based on small-volume urine sample - PubMed

  • Early detection of urothelial carcinoma (UCa) is challenging due to limitations of current diagnostic methods.
  • A novel urine-based H4C6/TWIST1 methylation assay was evaluated for diagnostic accuracy in UCa.
  • The study analyzed 743 urine samples (387 UCa patients and 356 controls) from four hospitals.
  • DNA was extracted from 3.5 mL of morning urine, and methylation status was assessed via quantitative PCR.
  • The assay showed high diagnostic accuracy: 93.02% sensitivity and 92.13% specificity.
  • Performance was strong for high-grade (97.64%) and muscle-invasive tumors (97.50%).
